摘要: The first step of herpesviruses virion assembly occurs in the nucleus. However, exact site where nucleocapsids are assembled, genome and inner tegument acquired, remains controversial. We created a recombinant VZV expressing ORF23 (homologous to HSV-1 VP26) fused eGFP dually fluorescent viruses with protein additionally red tag (ORF9, ORF21 ORF22 corresponding UL49, UL37 UL36). identified nuclear dense structures containing major capsid protein, scaffold maturing protease, as well ORF22. Correlative microscopy demonstrated that correspond aggregates time-lapse video imaging showed they appear prior accumulation cytoplasmic capsids, presumably undergoing secondary egress, highly dynamic. Our observations suggest these might represent area important for and/or maturation before budding at membrane.
