摘要: Relationships between mevalonate and DNA synthesis were explored in quiescent human fibroblasts stimulated with platelet-derived growth factor (PDGF). Studies of others have indicated that mevalonate, or a product other than cholesterol, is essential for replication. The present studies designed to determine whether there was critical time the cell cycle when necessary later occur. Compactin mevinolin, inhibitors hydroxymethylglutaryl CoA reductase, employed block both DNA. inhibited sharp peak which occurs cells 24 h after PDGF addition concentration-dependent manner. This suppression not prevented by low density lipoprotein but fully reversed mevalonate. inhibitor during interval 10-20 addition. Its presence only before 10 20 had no effect. Conversely, could overcome compactin period from Mevalonate When added mevinolin-blocked 10-15 PDGF, mevinolin 68% overcome; contrast, 20% reversal seen 15-20 h. Addition 2-h 10-12 overcame (assayed at h) 50%. results show period, several S phase, PDGF-stimulated require order proceed comprised approximately especially early part this interval.
