摘要: Pirfenidone is an anti-inflammatory and anti-fibrotic drug that has shown efficacy in lung kidney fibrosis. Because inflammation fibrosis have been linked to the progression of osteoarthritis, we investigated effects oral a mouse model cartilage injury, which results chronic joint-wide mice lack hyaluronan synthase 1 (Has1-/- ) comparison wild-type. Femoral was surgically injured wild-type Has1-/- mice, administered food starting after 3 days. At 4 weeks, reduced appearance, on micro-computed tomography, pitting subchondral bone at, cortical surrounding, site injury. This corresponded with reduction fibrotic tissue deposits as observed gross joint surface photography. resulted significant recovery trabecular parameters affected by injury although effect less pronounced. also increased Safranin-O staining growth plate sham operation both genotypes. Taken together expression selected extracellular matrix, inflammation, genes, these indicate may confer chondrogenic bone-protective effects, well-known occur earlier wound-healing response than time point examined this study. Further investigations identify specific cell populations signaling pathways are responsive warranted, other drugs encourage repair prevent post-traumatic osteoarthritis. © 2017 Orthopaedic Research Society.
