摘要: Recently purified and molecularly cloned monocyte chemotactic activating factor (MCAF) may play a major role in recruiting monocytes the inflammatory process. We examined effects of potent anti-inflammatory agent, dexamethasone (DXS), on production this factor. Over wide range concentrations (10(-5) to 10(-8) M), DXS inhibited MCAF at mRNA protein level human fibrosarcoma cell line, which was stimulated with either IL-1 or TNF-alpha. turn-over synthesized that showed decreased stability mRNA. Furthermore, addition actinomycin D cycloheximide abolished effect DXS, indicating de novo synthesis were required for In addition, nuclear run-off analysis revealed also transcription IL-1- TNF-activated genes. Therefore, both destabilization inhibition gene contribute decrease steady state by DXS.
