Peptide-Conjugated Gold Nanorods for Nuclear Targeting

摘要: Resonant electron oscillations on the surface of noble metal nanoparticles (Au, Ag, Cu) create plasmon resonance (SPR) that greatly enhances absorption and Rayleigh (Mie) scattering light by these particles. By adjusting size shape particles from spheres to rods, SPR can be tuned visible near-infrared region (NIR) where biologic tissues are relatively transparent. Further, gold nanorods enhance Raman adsorbed molecules. These unique properties make especially attractive as optical sensors for biological medical applications. In present work, covalently conjugated with a nuclear localization signal peptide through thioalkyl-triazole linker incubated an immortalized benign epithelial cell line oral cancer line. Dark field images demonstrate located in both cytoplasm nucleus lines. Single micro-Raman spectra reveal enhanced bands well molecules nucleus. difference between This work represents important step toward imaging Raman-based intracellular biosensing linked ligand-nanorod probes.
