Flavonoids exert distinct modulatory actions on cyclooxygenase 2 and NF-κB in an intestinal epithelial cell line (IEC18)

摘要: Background and purpose:  Cyclooxygenase 2 (COX-2) is involved in inflammatory bowel disease, but the effect of flavonoids at intestinal epithelial level unknown. We aimed to characterize structure-activity relationship nine selected on COX-2 expression cell (IEC)18 cells. also investigated signal transduction pathway(s) responsible for effects observed. Experimental approach:  Intestinal 18, a non-tumour line with phenotype, was used. measured by Western blot involvement NF-κB pathway assessed blot, pharmacological inhibition, luciferase reporter assays nuclear translocation experiments. Key results:  The depended experimental conditions tested [non-stimulated lipopolysaccharide (LPS)-stimulated]. Flavonoids caused an increase NF-κB-dependent gene transcription under basal conditions. Conversely, LPS stimulation increased, decreased or did not affect levels depending specific type. Variable were observed extracellular regulated kinase/p38/c-Jun N-terminal kinase phosphorylation p50/65 translocation. Conclusion implications:  balance interference IκB-α other signalling targets, therefore depends type flavonoids. This expected result different In general, may limit while favouring it when inflammation present.
