Pregnancy Associated Plasma Protein A2 (PAPP-A2) Affects Bone Size and Shape and Contributes to Natural Variation in Postnatal Growth in Mice

摘要: Pregnancy associated plasma protein A2 (PAPP-A2) is a protease of insulin-like growth factor binding 5 and receiving increasing attention for its roles in pregnancy postnatal growth. The goals the present study were to characterize effects PAPP-A2 deletion on bone size shape mice at 10 weeks age, determine whether Pappa2 gene responsible previously-identified quantitative trait locus (QTL) contributing natural variation mice. Mice homozygous constitutive lighter than wild-type littermates, had smaller mandible dimensions shorter skull, humerus, femur, tibia, pelvic girdle, tail bone. Furthermore, reduced lengths more would be expected due effect body mass. In addition size, deficiency also altered as assessed by geometric morphometrics. less deep mandibles, girdles with feminine shape. Using complementation test, we confirmed that QTL, demonstrating contributes If similar functional exists other species, this girdle might explain previously-reported associations between SNPs developmental dysplasia hip humans, birthing cattle.
