摘要: Recombinant protein production in plants such as corn is a promising means to generate high product yields at low comparable cost. The anti-EGFR monoclonal antibody C225, cetuximab, well-characterized receptor antagonist recently approved for the treatment of refractory colorectal cancer. We initiated study test and compare functional activity glycosylated aglycosylated C225 produced stable transgenic seed. Both antibodies were shown be functionally indistinguishable from mammalian-derived demonstrating high-affinity binding EGF receptor, blocking ligand-dependent signaling, inhibiting cell proliferation. In addition, consistent with both possessed strong anti-tumor vivo. Acute dose primate pharmacokinetic studies, however, revealed marked increase clearance antibody, while vivo kinetics similar cetuximab. This experimentation established that corn-derived possess efficacy mammalian culture-derived offer cost effective alternative large-scale culture production.
