Effects of recombinant human bone morphogenetic protein‐2 on human bone marrow cells cultured with various biomaterials

摘要: Recombinant human bone morphogenetic protein-2 (rhBMP-2) is known to induce orthotopic and ectopic formation in vivo. Several vitro studies using rat or mouse clonal cell lines have shown that rhBMP-2 may be involved the differentiation of osteoblasts from osteoblast precursor cells stromal marrow. However, there little information available about effects on cultured marrow cells. We investigated osteoblastic various biomaterials. Human were divided into fresh cells, fibroblast colony-forming units (cfu-F, precursors), The with without biomaterials, including titanium alloy, pure titanium, cobalt hydroxyapatite. It was found (500 ng/mL) significantly stimulated alkaline phosphatase production by cfu-F. when alloy only showed an increase after stimulation. Production osteocalcin, a marker mature osteoblasts, not any combinations tested. These findings suggest inducing rather than stimulating further osteoblasts. In addition, grafts cfu-F potential promote at sites nonunion as well around joint prostheses. © 1997 John Wiley & Sons, Inc.
