摘要: Background and Objective: Schistosoma japonicum is the causative agent of schistosomiasis in Philippines. Current diagnostics suffer from low sensitivity accuracy, hence an accurate reliable diagnosis essential for its prevention control. In this study, a PCR-based assay detection patient stool serum samples was developed. Methodology: Three candidate primer sets targeting mitochondrial genes COX3, NAD4, NAD5 were assessed. COX3 pair used rest study sensitivity, specificity, performance testing. Lastly, using compared to Kato-Katz circumoval precipitin test (COPT). Results: primers showed be suitable as sequencing analyses gave high similarities 96-98% S. , while NAD4 no similarity any organisms. The PCR-assay shown have limit 4 ng/ul DNA specific only japonicum. detected seven out ten -spiked samples. Comparative testing with COPT specificity 100% both samples, but formalin-fixed stored Conclusion: This developed sensitive detect human Our results suggest that PCR could useful fresh clinical can further improved reference improve other diagnostic assays schistosomiasis.
