摘要: Neocortical interactions with the dorsal striatum support many motor and executive functions, such underlying functional networks are particularly vulnerable to a variety of developmental, neurological, psychiatric brain disorders, including autism spectrum Parkinson's disease, Huntington's disease. Relatively little is known about development corticostriatal interactions, in particular, virtually nothing molecular mechanisms that control generation prefrontal cortex-striatal circuits. Here, we used regional cellular situ hybridization techniques coupled neuronal tract tracing show Cadherin-8 (Cdh8), homophilic adhesion protein encoded by gene associated disorders learning disability susceptibility, enriched within striatal projection neurons medial cortex medium spiny forming direct or indirect pathways. Developmental analysis quantitative real-time polymerase chain reaction western blot data Cdh8 expression peaks at P10, when cortical projections start form synapses striatum. High-resolution immunoelectron microscopy shows concentrated excitatory striatum, knockdown impairs dendritic arborization dendrite self-avoidance. Taken together, our findings indicate delineates developing circuits where it strong candidate for regulating normal projections, morphology, synapses.
