摘要: Plants have developed sophisticated signalling networks that are involved in mediating developmental transitions and environmental signals. Mitogen-activated protein kinases (MPKs) a class of proteins cellular processes help plants to detect initiate appropriate responses numerous development inputs. The microtubule cytoskeleton plays pivotal role plant morphogenesis, although the mechanisms regulate microtubule-associated functions cells not well understood. I investigated whether perturbations organization triggered by MICROTUBULE ORGANIZATION] temperature-sensitive mutant (mon-i) could lead altered transcriptional activity, with particular interest genes encoding signal transduction components. showed cytoskeleton, achieved through disruption phenotype mon-i, led changes expression gene transcripts associated diverse processes, including PROPYZAMIDE HYPERSENSITIVE 1(PHS1) gene, member MPK-specific pathway, which has been previously implicated cortical cells. Through biochemical, cell biological genetic tools, identified MPK1 8 as one MPKs interacts PHS 1 phosphatase demonstrated reverse genetics analysis manipulation MPK 18 results conditional subtle defects functionality. In contrast, 12, was shown also interact 1, no microtubule-specific function. My live imaging studies revealed absence appears effect on plus end growth shrinkage rates, indicating indirectly influences functions. Based analysis, MOR1 itself does appear be target putative module. Preliminary attempts obtain evidence for direct impacts activity failed demonstrate ofPHS1 either subcellular localization or phosphorylation status protein. These provide platform should facilitate future investigations aimed at understanding regulation
