摘要: Little is known regarding the mechanisms underlying infertility in male-to-female sex-reversed females. To gain a better understanding of germ cell dysfunction this condition, we produced XY sry− -females via CRISPR/Cas9 system C57BL/6 inbred strain mice. Mutant mice showed severe attrition cells during foetal development, resulting depletion ovarian before sexual maturation. Comprehensive transcriptome analysis embryonic day 13.5 primordial (PGCs) and postnatal 1 oocytes demonstrated that -PGCs had already deviated from developmental process at mitotic stage. In addition, -oocytes resulted meiotic disruption failure, genes related to these processes were significantly changed. This grievous disruption, which caused deterioration -females, was shown proceed themselves. These results provide novel insight into as well disorders sex differentiation human, such Swyer syndrome, wherein patients present typical females albeit with an karyotype infertile.
