Optimization of lipase activity from Rhizopus sp. in triglyceride hydrolysis using a modified simplex method

摘要: Abstract The simultaneous effect of reaction parameters and component concentrations on the microbial lipase from Rhizopus sp. was investigated. modified simplex optimization method applied to determine best operating conditions. activity measurement carried out with pH-stat where fatty acids liberated during enzymic hydrolysis olive oil substrate were titrated NaOH in an automatic tritrimeter. Two independent optimizations resulted a fully converged respect all parameters. Moreover, optimum parameter levels provided maximum activity, indicating that global has been found. 17·9 U/mg obtained at 40°C, pH 8·3, 41·8 mg/ml gum arabic, 0·6 sodium taurocholate. This response more than 100% better previous one by ‘one-factor-at-a-time’ method.
