摘要: Publisher Summary This chapter presents a method to calculate the apparent partial specific volumes of proteins in 6M Guanidine Hydrochloride (GuHCl), which greatly reduces uncertainties inherent previously used procedures. The only information required is amino acid composition protein. buoyancy sedimenting protein may be affected by GuHCl denaturation. It has been found that major change due interactions denatured with solvent components and volume upon denaturation 6 M generally small, its effect on being negligible. also calculation for α-chymotrypsin illustrate expressed as residues per 105 grams For glycoproteins, additional binding calculated assuming each residue carbohydrate binds one molecule GuHCl. In case coenzyme A transferase, total contents are about 1.6% weight. Therefore, final results essentially identical whether or not corrections carbohydrates made.
