摘要: Background & Aims Mice exposed to the hepatocellular carcinogen diethylnitrosamine at 2weeks of age have a high risk developing primary liver tumors later in life. Previous studies demonstrated that diethylnitrosamine-treated mice increased tumor burden when fed an obesigenic "Western" diet rich lard fat and sugar. However, role dietary fats vs. sugars promotion cancer is poorly understood. The aim this study was determine how altering affects model. Methods C57BL/6N were treated with and, from 6 32weeks age, one five diets differed sugar content, including normal chow, ketogenic, Western diets. Results sugar-rich had greatest irrespective content. In contrast, high-fat low-sugar least despite obesity glucose intolerance. When evaluated as independent variables, positively correlated hepatic accumulation, postprandial insulin, IL-6, inversely serum adiponectin. did not correlate adiposity, fasting or Furthermore, lower expression p21 cleaved caspase-3 compared low Conclusions These data indicate intake contributes excess adiposity insulin resistance diethylnitrosamine.
