摘要: Metallothionein (MT) is a potent hydroxyl radical scavenger but its antioxidant properties in vivo have not been defined. Most of the recent results indicate that it does afford protection to cells against lethal action oxidative stress. However, possibility MT confers damage specific cellular target, such as DNA, had considered. We compared V79 Chinese hamster enriched and depleted terms DNA-strand scission. Zinc induces an increase content cells, without concomitant GSH level. These induced are more resistant production scission by H2O2 than parental cells. Conversely, rendered partially deprived MT, transfection with plasmid vector which MT-I cDNA antisense oriented relation simian virus 40 promoter, became susceptible DNA-damaging H2O2. The transfected did exhibit alterations GSH, superoxide dismutase- H2O2-destroying enzymes. Indirect immunofluorescence indicated most was concentrated cell nucleus. Neither overexpression nor lower expression resulted differential resistance killing combined high nuclear concentration excellent confer DNA from attack.
