PDLIM5 identified by label-free quantitative proteomics as a potential novel biomarker of papillary thyroid carcinoma

摘要: In order to better understand the mechanisms underlying development of papillary thyroid carcinoma (PTC), and identify new potential biomarkers, high-resolution label-free mass spectrometry was performed on PTC tissues adjacent normal from six patients. this process, 2788 proteins were identified, out which 49 presented significant differences between tissues. Gene ontology revealed that majority these are involved in catalytic activity binding. We selected three with differential expressions: PDZ LIM domain 5 (PDLIM5), PDLIM1 ALDH1A1; Protein expressions further verified by RT-PCR western blot. Among these, expression PDLIM5 up-regulated, while ALDH1A1 down-regulated Next, we confirmed their through quantitative dot blot (QDB) technique. found knockdown B-CPAP cell line could inhibit migration, invasion proliferation cells. addition, reduced Ras Phospho-ERK1/2 expression. Thus, suggested promotes via activation Ras-ERK pathway. Our research provides molecular insight into function PDLIM5, may assist studying mechanism PTC. be explored as a candidate for treatment.
