摘要: We describe a technological approach permitting the massive expansion of CD34(+) stem cells and their 100% conversion ex vivo into mature red blood (RBC). The protocol comprises three steps: first step consisting cell proliferation induction erythroid differentiation in liquid medium without serum presence growth factors (GF), second based on model reconstitution medullar microenvironment (ME) (human MSC or murine stromal cells) GF, third ME alone, any GF. This work highlights impact terminal maturation cells. A critical point is that RBC generated vitro have all characteristics functional native adult RBC. Moreover, this new concept 'cultured RBC' (cRBC) important for basic research erythropoiesis has major clinical implications, especially transfusion medicine. three-step can be adapted to use hematopoietic (HSC) from diverse sources: peripheral blood, bone marrow cord blood.
