摘要: Matrix metalloproteinases also known as MMPs are zinc-dependent endoproteases which process extracellular matrix proteins at neutral pH to regulate normal macromolecular protein turnover well those cellular events associated with the remodeling of tissue architecture. The MMP superfamily include classical MMPs, MMP-1, -8, -13, and -18, gelatinases, MMP-2, -9, stromelysins, MMP-3, -10, -11, matrilysins, MMP-7, MMP-26, membrane-type (MT-MMPs; MMP-14, -17, 24/25) ADAMS (a disintegrin metalloproteinase) adamlysins ADAMTS metalloproteinase thrombospondin motif). genes regulated principally via activation transcription factors induced by various growth cytokine/cytokine receptor interactions pertinent allostasis, but most critical pathophysiological progression rheumatoid arthritis osteoarthritis. important be involved in regulating gene expression synthesis and/or NF-κB, Cbfa1, AP-1, Nmp4/CIZ, ELF3, c-Maf, KLF5 Sp1 interact specific sequences promoter region genes. In arthritis, kinase pathways generally activated pro-inflammatory cytokines, including interleukin-1β, tumor necrosis factor-α, interleukin-6, other members IL-6 family. these signaling mechanisms have traditionally been considered “undruggable.” However, recent experimental evidence indicates that inhibition factor may achieved could employed suppress activity.
