摘要: Plasma nicotine concentrations following administration by two types of nasal spray were compared in ten subjects. Absorption was particularly rapid during the first 2.5 min, average rise blood this time being 8.6 ng/ml for products, followed a small further to an peak increase 10.5 5 min after dose 2 mg base (mean 27.8 µg/kg). Despite four-fold Cmax variation between subjects, levels individual subjects fairly consistent across products. There no significant differences products or cardiovascular responses, and correlation AUCs from 0.68 (P=0.01). Eight reported subjective feelings light-headedness slight dizziness, which are not typical slower absorption gum skin patches. Blood within smoking range soon built up with repeated doses, even subject least efficient absorption. In second study ad libitum use under clinical conditions both appeared sufficiently acceptable therapeutic as aid cessation. tendency escalate excessive over 4 weeks, nine averaged only 44% their prior levels. Only one had equivalent possible reasons why more common discussed.
