摘要: One of the key enzymes in chemolithoautotrophic metabolism Alcaligenes eutrophus H16 is a dimeric, membrane-associated hydrogenase. The genetic determinants this enzyme are located on endogenous megaplasmid pHG1 (G. Eberz, C. Hogrefe, Kortluke, A. Kamienski, and B. Friedrich, J. Bacteriol. 168:636-641, 1986). Complementation studies showed that information required for formation active membrane-bound hydrogenase occupies more than 7.5 kb DNA. We cloned sequenced region identified genes encoding two subunits (hoxK hoxG). nucleotide sequence contains nine additional closely spaced open reading frames. Immunoelectron microscopy gene product one these frames (hoxM) involved process leading to attachment membrane. Other may encode processing functions components hydrogenase-linked electron transport chain. Analysis Tn5-B21-mediated transcriptional fusions provided evidence structural accessory belong at least three coordinately regulated units.
