A putative ATP-activated Na+ channel involved in sperm-induced fertilization

摘要: Extracellular application of adenosine triphosphate (ATP) to defolliculated Xenopus laevis oocytes activated a saturating inward current with maximal amplitude E(max) 2.4 +/- 0.2 microamperes and an apparent Michaelis constant 197.6 micromolar. The was carried predominantly by sodium ions potently inhibited amiloride, guanosine (GTP), its nonhydrolyzable analogs 5'-[beta,gamma-imido]triphosphate (GppNHp) 5'-O-(3-thiotriphosphate). Likewise, in vitro fertilization using mature eggs sperm GTP, GppNHp. Hence, ATP receptor on the egg membrane may be recipient target for originating sperm, suggesting that ATP-induced increase permeability mediates initial signal process.
