摘要: Background Histamine-induced wheal-and-flare studies are useful, objective tests for determining differences in the peripheral H 1 -receptor blockade activities of antihistamines. Objective To evaluate time occurrence 95% inhibition histamine-induced wheal and flare after administration fexofenadine hydrochloride, 180 mg, or cetirizine, 10 mg. Methods Forty-two volunteers (aged 18–60 years) were included a randomized, double-blind, crossover study. Skin prick undertaken using histamine (100 mg/mL) before treatment 0.5, 1.0, 1.5, 2.0, 2.5, 3.0, 3.5, 4.0 hours treatment. Wheal areas evaluated, to frequency subjects exhibiting median calculated. Results Mean ± SD was 2.46 0.71 with 2.55 0.57 cetirizine. The estimated mean difference between cetirizine (−7 minutes favor fexofenadine; 2-sided confidence interval, −21 +7 minutes) not statistically significant ( P = .34). For wheal, 29% receiving 24% achieved (2.5 hours). An exact permutation test yielded .37. flare, 26% 10% (3 hours; .12 by test). Conclusions Fexofenadine have comparable onset action times similar frequencies inhibition, as evaluated flare.
