Direct Identification of Bacteria in Positive Blood Culture Bottles by Matrix-Assisted Laser Desorption Ionisation Time-of-Flight Mass Spectrometry

摘要: Background With long delays observed between sampling and availability of results, the usefulness blood cultures in context emergency infectious diseases has recently been questioned. Among methods that allow quicker bacterial identification from growing colonies, matrix-assisted laser desorption ionisation time-of-flight (MALDI-TOF) mass spectrometry was demonstrated to accurately identify bacteria routinely isolated a clinical biology laboratory. In order speed up process, present work we attempted directly culture bottles detected positive by automate. Methodology/Principal Findings We prospectively analysed routine MALDI-TOF two different protocols involving successive centrifugations then lysis trifluoroacetic acid or formic acid. Of 562 broths as automate containing one species, 370 (66%) were correctly identified. Changing protocol improved Staphylococci, overall correct increased 59% 76%. Lack mostly with viridans streptococci, only false observed. 22 contained more species identified 18 samples, no samples identifications obtained cases. The predictive value using this procedure 99.2%. Conclusions/Significance MALDI-TOF MS is an efficient method for direct isolates culture, exception polymicrobial streptococci. It may replace performed on provided improvement specificity streptococci near future.
