摘要: Spt6 is a conserved factor, critically required for several transcription- and chromatin-related processes. We now show that its binding partner, Iws1, are heterochromatic silencing in Schizosaccharomyces pombe. Our studies demonstrate of all loci an spt6 mutant has unusual combination phenotypes compared to previously studied mutants. Unexpectedly, we find normal nucleosome positioning over heterochromatin levels histone H3K9 dimethylation at the endogenous pericentric repeats. However, also greatly reduced trimethylation, elevated H3K14 acetylation, recruitment factors, defects spreading. evidence suggests plays role both transcriptional posttranscriptional levels; mutant, RNA polymerase II (RNAPII) occupancy regions only modestly increased, while production small interfering RNAs (siRNAs) lost. Taken together, our results suggest multiple steps by controlling chromatin, transcriptional, from prokaryotes (49) humans (25) many roles regulation transcription chromatin structure. Studies organisms, particularly Saccharomyces cerevisiae, have elucidated functions.
