摘要: Intracellular and extracellular sources of cytosolic [Ca2+] elevation in isolated rat osteoclasts were explored by a comparison fura-2 signals response to application the Ca2+ ionophore, ionomycin, Ca(2+)-containing Ca(2+)-free bathing solutions. Cytosolic transients persisted bathed Ca(2+)-free, EGTA-containing They consisted peak [Ca2+bd followed full decay baseline refractory manipulations surface membrane potential through changes [K+]. disappeared upon intracellular store depletion repeated ionophore applications. therefore attributable solely intracellularly stored Ca2+. In contrast, peaks exposed solutions decayed sustained levels. responses then with ionomycin application. These latter phenomena are accordingly entry. Finally, restoration 1.25 mM following stores treatment elicited 'overshoot' consistent capacitative entry via route. results demonstrate refillable source that could function osteoclastic regulation.
