Intranasal delivery to the central nervous system: Mechanisms and experimental considerations

摘要: The blood-brain barrier (BBB) limits the distribution of systemically administered therapeutics to central nervous system (CNS), posing a significant challenge drug development efforts treat neurological and psychiatric diseases disorders. Intranasal delivery is noninvasive convenient method that rapidly targets CNS, bypassing BBB minimizing systemic exposure. This review focuses on current understanding mechanisms underlying intranasal with discussion pathways from nasal cavity CNS involving olfactory trigeminal nerves, vasculature, cere- brospinal fluid, lymphatic system. In addition properties thera- peutic, deposition formulation within passages composition can influence pathway therapeutic follows into after administration. Experimental factors, such as head position, volume, administration, parameters, pH, osmolarity, or inclusion permeation enhancers mucoadhesives, formula- tion followed CNS.
