摘要: Background Hemophilic arthropathy is a debilitating morbidity of hemophilia caused by recurrent joint bleeds. We investigated if the bleed volume, before initiation treatment, was linked to subsequent degree histopathological changes and development bone pathology in mouse model hemophilic arthropathy. Methods FVIII knock-out (F8-KO) mice were dosed with micro-CT blood pool agent prior induction hemarthrosis. Eight hours after induction, volume quantified micro computed tomography (micro-CT) recombinant treatment initiated. On Day 8, inflammation knees characterized fluorescence molecular tomography. 14, knee histopathology. In second study, contrast injected into wild-type (WT) mice, followed evaluation on 14. Results The average 3.9 mm3. inflammation-related fluorescent intensities injured significantly increased 8. had synovitis scores, vessel counts, areas hemosiderin compared un-injured knees. However, no cartilage- or observed. correlated associated high intensity F8-KO WT persistence elicited morphological changes. Conclusion When applying delayed on-demand regimen subjected an induced hemarthrosis, 14 reflected treatment.
