摘要: A 5417-base pair (bp) region of Pseudomonas aeruginosa PAO chromosomal DNA containing the mmsAB operon and an upstream regulatory gene (mmsR) has been cloned characterized. The contains two structural genes involved in valine metabolism: mmsA, which encodes methylmalonate-semialdehyde dehydrogenase; mmsB, 3-hydroxyisobutyrate dehydrogenase. mmsA mmsB share same orientation are separated by a 16-bp noncoding region. transcriptional start site for pinpointed to cytidine residue located 77 bp from translational operon. mmsR is on opposite strand begins 134 mmsA. MmsR identified as member XylS/AraC family regulators appears act positive regulator Sequence comparison MmsA other proteins data bases revealed that belongs aldehyde dehydrogenase (NAD+) superfamily. MmsB shares 44% amino acid identity with rat liver. Mutants insertionally inactivated mmsR, grow slowly valine/isoleucine medium exhibit reduced enzyme activity cell-free extracts compared P. PAO.
