摘要: Summary Recent studies of Mn2+ transport mutants indicate that manganese is essential for unstressed growth in some bacterial species, but required primarily induced stress responses others. A Bradyrhizobium japonicum mutant defective the high-affinity transporter gene mntH has a severe phenotype under limitation, suggesting requirement metal growth. Here, we found activities superoxide dismutase and glycolytic enzyme pyruvate kinase were deficient an strain grown limitation. We identified pykM as only kinase-encoding based on deficiency activity mutant, rescue with pyruvate, purified recombinant PykM. PykM unusual it rather than Mg2+ high activity, neither fructose-1,6-bisphosphate nor AMP was positive allosteric effector. The mntH-dependent encoded by sodM, expressed dismutase-encoding conditions. An grew more slowly manganese-limited conditions did sodM double implying additional manganese-dependent processes. findings implicate roles key steps oxidative metabolism B. japonicum.
