摘要: Abstract The 3' untranslated region (3'-UTR) has been implicated in the estrogen stabilization of hepatic Xenopus laevis vitellogenin mRNA. We used RNA gel mobility shift assays to demonstrate that liver contains a factor which binds with very high specificity segment 3'-UTR B1 and B2 mRNAs. detected single high-affinity binding site mRNA localized 27-nucleotide region. Since was abolished by proteinase K digestion, at least component is protein. Following administration, induced approximately four- fivefold extracts from polysomes. The mRNA-binding protein found both polysomes cytosol. also inducible cytosol, this represents genuine induction, not simply recruitment cytosolic into UV cross-linking studies recognition sequence revealed bands corresponding bound proteins apparent molecular weights 71,000 141,000. This appears be first example steroid hormone-inducible an 3'-UTR. Its induction its sequence-specific important estrogen-mediated suggest may play role regulation stability.
