摘要: We cloned a DNA fragment directing synthesis of A-factor from the total cellular streptomycin-producing Streptomyces bikiniensis on plasmid vector pIJ385 . Introduction recombinant ( pAFB1 ) into A-factor-deficient S. and griseus mutants led to production in host cells, as result which streptomycin production, resistance, spore formation these were simultaneously restored. The also complemented both afsA afsB mutations coelicolor A3(2). These results indicated that contained genetic determinant biosynthesis. fragment, when carried multicopy plasmid, induced large amount several hosts. In Southern blot DNA/DNA hybridization analyses with trimmed 5-kilobase containing intact probe, gave no positive hybridization. experiment showed wide distribution sequences homologous among most, but not all, A-factor-producing actinomycetes varying extent homology absence most nonproducers Images
