摘要: Mechanism of fibronectin (FN)-induced chemotaxis fibroblastic cells has not been fully understood. The present study was performed to establish a molecular nature the chemotactic region rat plasma FN. dose-response pattern intact FN for mouse embryo cells, NIH-L13 which represented as "bell-shape" curve with maximum activity at around 50 nM, changed "biphasic" mode through proteolysis thermolysin. Two distinct components were isolated from thermolytic fragments. One component, fragment mass 110-150 kDa, estimated contain central cell-binding domain and carboxyl-terminal heparin-binding molecule. Cell migration stimulated by 110-150-kDa increased successively in dose-dependent manner, capability promote much higher than that (over 2-fold). second 21 shown reside fibrin-binding domain. 21-kDa produced bell-shape pattern, being consistent FN, whereas response occurred 100-fold lower concentration (0.5 nM) At concentrations, this revealed an inhibitory cell fragment. No significant interaction between these two active observed polyacrylamide gel electrophoresis under nondenaturing conditions, suggesting may act directly on inhibit migration. These results suggest contains least chemotactically regulate cooperatively cells.
