摘要: Murine monoclonal antibody 196-14 recognizes the ovarian-cancer-associated antigen CA 125, but epitope it is different from that of OC125. We developed a human/mouse chimeric using variable regions murine and human heavy-chain (γl) light-chain (κ) constant regions. Cell binding competitive inhibition assays labeled with125I,111In or99mTc demonstrated in vitro immunoreactivity was identical to parental antibody. However, mice bearing ovarian cancer xenografts, clearance blood faster absolute levels accumulation tumor were lower for the125I-labeled or99mTc-labeled than with corresponding radionuclides. The tumor-to-blood radioactivity ratio not significantly between antibody, regardless radionuclide used labeling. Chimeric with131I,111In promising radioimmunoimaging cancer.
