Regulation the morphology of cationized gold nanoparticles for effective gene delivery

摘要: Recent research indicated that the morphology of nanoparticles could result in distinct biological behaviors, thus played an important role designing efficient gene delivery systems. Among them, gold (AuNPs) with various shapes were widely studied due to good biocompatibility and easy modification ability. Our recent polyethyleneimine-g-bovine serum albumin (BSA-PEI) as non-viral vector showed colloid stability high transfection efficiency. In this work, BSA-PEI was utilized modify nanospheres (AuNSs) nanorods (AuNRs) investigate influence on delivery. Both AuNS@BSA-PEI AuNR@BSA-PEI condensed DNA effectively at N/P ratio above 5 maintained spherical or rod-like respectively. Due higher surface charge density tips, complexes prone use tips contact cell membrane, which facilitated be uptaked by HepG2 cells. The endocytosis inhibition experiments some differences endocytic pathway. Gene experiment had almost 100-fold level than 20. This work provided a potential strategy for further design vectors improved results adjusting vectors.
