摘要: Standard Ginkgo biloba leaf extract (EGb 761) has been known to have neuroprotective effects ranging from molecular and cellular, animal human studies, however, the cellular mechanisms remain unclear. Using PC 12 cells, a well-established model for studying neuroprotection, we determined mechanism of action EGb 761 on cell survival following apoptosis induced by serum-deprivation or treatment with staurosporine (STS). Our results show that treatments PC12 cells are able prevent serum deprivation- STS-induced mitochondrial damage, attenuate release cytochrome c DNA fragmentation. 761, but not vitamin E. inhibited activation caspase-3 enzyme. Two components, bilobalide B ginkgolide C more significant inhibition than extract. Furthermore, microarray assay indicate transcription multiple apoptosis-related genes is either up- down-regulated in treated 761. These suggest apoptotic machinery may, at least part, mediate E act different paths provide neuroprotection.
