Imaging heterogeneity in the mitochondrial redox state of premalignant pancreas in the pancreas-specific PTEN-null transgenic mouse model

摘要: Metabolic alteration is one of the hallmarks carcinogenesis. We aimed to identify certain metabolic biomarkers for early detection pancreatic cancer (PC) using transgenic PTEN-null mouse model. Pancreas-specific deletion PTEN in caused progressive premalignant lesions such as highly proliferative ductal metaplasia. imaged mitochondrial redox state pancreases mice approximately eight months old scanner, i.e., nicotinamide adenine dinucleotide/oxidized flavoproteins (NADH/Fp) fluorescence imager at low temperature. Two different approaches, global averaging indices without considering tissue heterogeneity along depth and univariate analysis multi-section data a covariate were adopted statistical imaging data. The standard deviations histogram with Gaussian fit used determine heterogeneity. All methods show consistently that deficient (Pdx1-Cre;PTENlox/lox) significantly more heterogeneous their compared controls (PTENlox/lox). Statistical taking into account variations further shows shifted an overall oxidized state. Oxidization group was not seen when analyzed by variation depth, indicating importance This study reveals possible link between pancreas its malignant transformation may be developed establishing potential diagnosis cancer.
