摘要: Nucleic acid sample preparation has been an especially challenging barrier to point-of-care nucleic amplification tests in low-resource settings. Here we provide a head-to-head comparison of methods for lysis of, and release from, several pathogenic bacteria viruses-methods that are adaptable usage Digestion with achromopeptidase, mixture proteases peptidoglycan-specific hydrolases, followed by thermal deactivation boiling water bath, effectively released amplifiable from Staphylococcus aureus, Bordetella pertussis, respiratory syncytial virus, influenza virus. Achromopeptidase was functional after dehydration reconstitution, even eleven months dry storage without refrigeration. Mechanical proved be effective against hard-to-lyse Mycobacterium species, miniature bead-mill, the AudioLyse, is shown capable releasing DNA RNA this species. We conclude point-of-care-compatible need not introduce inhibitors, can amplification-ready lysates wide range bacterial viral pathogens.
