摘要: Direct reprogramming of somatic cells to induced pluripotent stem (iPSCs) provides an invaluable resource for regenerative medicine. Because some ethical and logistical barriers, human iPSCs cannot be used generate a chimera, which is one markers representing pluripotency. As the most attractive model preclinical studies, pigs offer another path improve clinical In this study, porcine adult (pASCs), including adipose mesenchymal (AMSCs) bone marrow (BMSCs), were collected cultured under same conditions in vitro. Real-time PCR, immunocytochemical staining, apoptosis analysis, differentiation techniques investigate proliferative capacity characteristics pASCs. Our results showed that both AMSCs BMSCs displayed similar immunophenotype, their appeared as downward trend cell passage number increased. The was significantly lower than (p<0.05). Moreover, each type pASCs went through 20 passages without undergoing alterations expression transcriptional factors (Oct4, Sox2, c-Myc, Nanog). All had adipogenic osteogenic potential. addition, they also could reprogrammed pig (piPSCs) with time efficiency. conclusion, higher AMSCs, pluripotency stable long-term culture.
