Effects of Hypocretin/Orexin and Major Transmitters of Arousal on Fast Spiking Neurons in Mouse Cortical Layer 6B

摘要: Fast spiking (FS) GABAergic neurons are thought to be involved in the generation of high-frequency cortical rhythms during waking state. We previously showed that layer 6b (L6b) was a specific target for wake-promoting transmitter, hypocretin/orexin (hcrt/orx). Here, we have investigated whether L6b FS cells were sensitive hcrt/orx and other transmitters associated with activation. Recordings thus made from either wild-type mice or transgenic which GFP-positive parvalbumin positive. Whereas control condition induced strong increase frequency, but not amplitude, spontaneous synaptic currents, presence TTX, it had no effect at all on miniature currents. Hcrt/orx presynaptic although by an action glutamatergic terminals rather neighboring cells. In contrast, noradrenaline acetylcholine depolarized excited these through direct postsynaptic action. Neurotensin, is colocalized neurons, also indirect. Morphologically, exhibited basket-like features. These results suggest hcrt/orx, noradrenaline, acetylcholine, neurotensin could contribute activity
