摘要: Abstract Hyperglycemia induces apoptotic cell death in a variety of types diabetes, and the mechanism remains unclear. We report here that culture rat retinal glial Muller cells 25 mM glucose for 72 h significantly inactivated Akt induced apoptosis. Likewise, hyperglycemia caused significant dephosphorylation at serine-473 retina streptozotocin-induced diabetic rats. Inactivation was associated with BAD, increased cytochrome c release, activation caspase-3 caspase-9. Upregulation activity by overexpression constitutively active inhibited elevated glucose-induced apoptosis, whereas downregulation dominant negative exacerbated as assessed caspase nucleic acid staining. These data suggest apoptosis chronically is least part mediated survival pathway cultured cells. It has been reported antiapoptotic effect requires growth factor withdrawal-induced Our although acutely may be beneficial to survival, can cause via signaling.
