Differential Modulation of Protein Kinase C Isoforms in Erythroleukemia during Induced Differentiation

摘要: Induction of erythroid differentiation murine erythroleukemia cells (MELC) by exposure to hexamethylene bisacetamide (HMBA) involves the modulation protein kinase C (PKC) activity. Using immuno- and Northern blot techniques, we have demonstrated that MELC express a pattern PKC isoforms which includes alpha, delta, epsilon, zeta, eta. We show resistant induction HMBA significantly less nPKC isoform, slightly epsilon. Recovery sensitivity is associated with reexpression delta protein. Upon HMBA, there fall in cytosolic epsilon accompanied transient increase membrane-associated forms these isoforms. HMBA-resistant fail display this isoform-specific translocation PKC. accompanied, over next 24 h progressive cellular activity, content zeta. These studies suggest possibly zeta as well, play role pathway HMBA-mediated terminal cell MELC.
