摘要: Autosomal dominant polycystic kidney disease (ADPKD) is characterized by progressive enlargement of cysts, leading to chronic disease. Since the available treatment for ADPKD limited, there emerging interest natural compounds as potential therapeutic candidates. The aim our study was investigate whether an olive leaf extract may be able counteract cyst growth in vitro model ADPKD. We treated WT9-12 cells with (OLE). In monolayer culture we evaluated cell viability MTT assay, protein expression western-blot analysis and apoptosis DNA laddering TUNEL assays. For functional studies used transient transfection ChIP Intracellular calcium measurement performed a spectrofluorimeter using fluorescent probe. 3D-cell-culture studies. OLE reduced rate affected intracellular signaling due high c-AMP levels, PKA enhanced p-AKT, restored B-Raf-inactivation down-regulated p-ERK. elucidated molecular mechanism which OLE, via Sp1, transactivates p21WAF1/Cip1 promoter, whose levels are mutated PKD1. demonstrated that p-AKT up-regulation also played crucial role OLE-induced anti-apoptotic effect ameliorated primary cause Finally, observed size.
