Identification of Suppressors of mbk-2/DYRK by Whole-Genome Sequencing

摘要: Screening for suppressor mutations is a powerful method to isolate genes that function in common pathway or process. Because often do not have phenotypes on their own, cloning of loci can be challenging. A combining whole-genome sequencing (WGS) and single nucleotide polymorphism (SNP) mapping (WGS/SNP mapping) was developed identify with visible C. elegans. We show here WGS/SNP an efficient map without the need previous phenotypic characterization. Using RNA-mediated interference test candidate identified by mapping, we 10 extragenic six intragenic suppressors mbk-2, DYRK family kinase required transition from oocyte zygote. Remarkably, seven are cell-cycle regulators extend timing oocyte-to-zygote transition.
