摘要: The human, murine, and rat B29 (Igβ, CD79b) genes are highly conserved in sequence organization exhibit strict B cell-specific expression. In the human genomes, gene is located between skeletal muscle-specific Na-channel αsubunit (SCN4A) pituitary-specific growth hormone (GH-N) gene. GH-N controlled by a tissue-specific locus control region (LCR) just upstream of promoter that mediates enhancement, histone acetylation, an open chromatin conformation across (GH)-expressing pituitary cells. Here we show mRNA not detected GH-expressing cell line This differential expression suggests insulated or otherwise protected from regulatory influences closely proximal GH LCR. We searched available sequences mouse, found fulfills criteria recently established for non-coding DNA elements potentially involved control. (BCS) bound ubiquitously expressed nuclear protein complexes. DNase I protection analysis BCS revealed central `footprinted' core which was confirmed to bind multifunctional transcription factor, YY1. However, neither nor YY1-binding motif exhibited silencer enhancer activity transient transfections position-independent insulator enhancer-blocking assays.
