Examining Histone Posttranslational Modification Patterns by High-Resolution Mass Spectrometry

摘要: Histone variants and posttranslational modifications (PTMs) are essential for epigenetic regulation of transcriptional expression. Single and/or combinatorial PTMs histones play important roles in development disease formation. Mass spectrometry (MS) has been a powerful tool to study histone PTMs, as it not only can identify novel but also provide quantitative measurement spectrum the same sample. In this chapter, we employ combination chemical derivation high-resolution MS quantify multiple PTMs. Histones acid extracted modified with propionyl groups subsequently produces suitable sizes fragments analysis by trypsin digestion. The newly generated N-termini peptides be differentially marked stable isotope labeling second reaction propionylation, which enables direct comparison between two different samples following analysis.
