A variant of the gene encoding leukotriene A4 hydrolase confers ethnicity-specific risk of myocardial infarction

摘要: Variants of the gene ALOX5AP (also known as FLAP) encoding arachidonate 5-lipoxygenase activating protein are to be associated with risk myocardial infarction 1 . Here we show that a haplotype (HapK) spanning LTA4H leukotriene A4 hydrolase, in same biochemical pathway ALOX5AP, confers modest an Icelandic cohort. Measurements B4 (LTB4) production suggest this is mediated through upregulation pathway. Three cohorts from United States also HapK relative (1.16) European Americans, but it threefold larger African Americans. About 27% American controls carried at least one copy HapK, compared only 6% controls. Our analyses indicate very rare Africa and its occurrence Americans due admixture. Interactions other genetic or environmental factors more common likely account for greater conferred by group.
