摘要: Publisher Summary This chapter discusses the isolation and characterization of temperature-sensitive mammalian cell cycle mutants. Except for sex chromosomes, cells have a diploid set chromosomes. Therefore, it is difficult to isolate recessive mutant from cells. Furthermore, cultured lines are an artificial biological material, since they derived tissues. When cut out tissues plated on dishes, initially grow well. Cell presently survived such crisis acquired ability dishes permanently, if materials necessary proliferation supplied. To characterize ts mutants at molecular level, essential identify genes that cause phenotype. analysis has been improved by combining synchronization with expression pattern cycle-specific genes.
