RADYASYONUN SERUM DEMİR, BAKIR VE PROTEİN OKSİDASYON DÜZEYLERİNE ETKİSİ

摘要: Amac: Bir cok hastaligin etyopatogenezinde rol oynadigi dusunulen prote- inlerin oksidatif modifikasyonunu analiz etmek amaciyla en sik kullanilan yontem protein oksidasyon urunu olan karbonil gruplarinin tayinidir. Biz de calismamizda, radyasyonun serum uzerindeki etkisini gostermek grubu duzeylerini ve “metallerin katalizledigi oksidasyon” (metal-catalyzed oxidation, MCO) sistemleri ara- ciligiyla oksidasyonunu artirdigi bilinen demir (Fe) bakir (Cu) arastirmayi amacladik. Gerec Yontemler: Kobaylara radyasyon antioksidan (N-asetilsistein, NAC) uygulamalari sonrasinda Fe, Cu duzey- leri edildi. Protein duzeyi Levine metoduna gore, Fe du- zeyi otoanalizorde, ise atomik absorbsiyon cihazinda olculdu. Bulgular: Radyasyonun, grubu, duzeyle- rinde istatiksel olarak anlamli artislara neden oldugu goruldu. Radyasyon oncesinde NAC verilen grupta tek basina uygulanan gruba oranla stresin azaldigi gozlendi. yalnizca veril- mesi parametrelerde azalmaya oldu. Sonuc: Sonuc muhtemelen reaktif oksijen turlerinin zeyini artirarak, ayrica yukseltip metal katalizli oksi- dasyon yoluyla proteinlerin modifikasyonuna yol actigi, NAC’in olusturdugu oksidasyona karsi koruyucu bir etkisinin ol- dugu tespit Anahtar Kelimeler: Radyasyon, oksidasyonu, grubu.
